Root-secreted nucleosides: signaling chemoattractants of rhizosphere bacteria

The rhizosphere is a complex ecosystem, consisting of a narrow soil zone influenced by plant roots and inhabited by soil-borne microorganisms. Plants actively shape the rhizosphere microbiome through root exudates. Some metabolites are signaling molecules specifically functioning as chemoattractants rather than nutrients. These elusive signaling molecules have been sought for several decades, and yet little progress has been made. Root-secreted nucleosides and deoxynucleosides were detected in exudates of various plants by targeted ultra-performance liquid chromatography–mass spectrometry/mass spectrometry. Rhizobacteria were isolated from the roots of Helianthemum sessiliflorum carrying the mycorrhizal desert truffle Terfezia boudieri. Chemotaxis was determined by a glass capillary assay or plate assays on semisolid agar and through a soil plate assay. Nucleosides were identified in root exudates of plants that inhabit diverse ecological niches. Nucleosides induced positive chemotaxis in plant beneficial bacteria Bacillus pumilus, Bacillus subtilis, Pseudomonas turukhanskensis spp., Serratia marcescens, and the pathogenic rhizobacterium Xanthomonas campestris and E coli. In a soil plate assay, nucleosides diffused to substantial distances and evoked chemotaxis under conditions as close as possible to natural environments. This study implies that root-secreted nucleosides are involved in the assembly of the rhizosphere bacterial community by inducing chemotaxis toward plant roots. In animals, nucleoside secretion known as “purinergic signaling” is involved in communication between cells, physiological processes, diseases, phagocytic cell migration, and bacterial activity. The coliform bacterium E. coli that inhabits the lower intestine of warm-blooded organisms also attracted to nucleosides, implying that nucleosides may serve as a common signal for bacterial species inhabiting distinct habitats. Taken together, all these may indicate that chemotaxis signaling by nucleosides is a conserved universal mechanism that encompasses living kingdoms and environments and should be given further attention in plant rhizosphere microbiome research.

AMF symbiosis drives the rhizosphere microbiome to synergistically improve herbage growth in saline–alkaline soils

AbstractPlant–microbe interactions are essential in shaping plant performance and overall ecosystem functioning. However, the regulatory mechanisms underlying plant–microbe interactions mediated by mycorrhizal symbiosis in saline–alkaline soils are still not fully understood. Here, we aimed to clarify the synergistic regulatory mechanism through which arbuscular mycorrhizal fungi (AMF) symbiosis drives the rhizosphere microbiome to improve perennial herbage growth in saline–alkaline soils and evaluate phytoremediation efficiency. This study revealed that Funneliformis mosseae inoculation (i) strongly promoted the growth of all three herbage species (with values ranging from 21.62% to 233.33%), Na+ accumulation in plants (with values ranging from 24.63% to 188.89%), and decreased soil electrical conductivity (with values ranging from 7.68% to 12.87%), potentially suggesting improved phytoremediation efficiency with AMF symbiosis; (ii) increased nutritional content and decreased C:P and N:P ratios (with values ranging from 27.20% to 92.87%) and improved K+/Na+ and P/Na+ ratios (with values ranging from 2.60% to 302.96%); (iii) increased the abundance of some beneficial bacterial taxa and strengthened the significant strong relationships among most of these bacteria and plant biomass, ion homeostasis as well as stoichiometric ratio constants, and AMF inoculation treatments also consisted the higher proportion of differential genera significantly correlated with these plant factors as well as plant nutrient contents, potentially reflecting that AMF mediated the enrichment process of beneficial bacterial taxa and may strength functional interaction between plant and bacterial taxa, which may be importance for the enhancement of saline–alkaline tolerance of plants; and (iv) enhanced stability of the rhizosphere bacterial community and complexity of interaction networks, and the related indictors also established significant correlations with plant/soil factors, suggesting that the improvement of stability and functional complexity driven by AMF may also be beneficial for enhancing phytoremediation efficiency. These findings indicate that AMF inoculation plays its own beneficial role by simultaneously activating the potential of beneficial rhizosphere bacterial taxa and that their synergistic interaction is more beneficial for enhancing plant growth in salt‐affected soils and enhancing phytoremediation efficiency. This study helps to elucidate the underlying mechanisms through which AMF‐mediated rhizosphere bacterial community improve plant growth and tolerance to saline–alkaline stresses, and provides evidence that effective ecological restoration of saline–alkaline degraded grasslands can be achieved via the use of mycorrhizal symbiosis herbage.

Bt-Modified Transgenic Rice May Shift the Composition and Diversity of Rhizosphere Microbiota

Plants significantly shape root-associated microbiota, making rhizosphere microbes useful environmental indicator organisms for safety assessment. Here, we report the pyrosequencing of the bacterial 16S ribosomal RNA in rhizosphere soil samples collected from transgenic cry1Ab/cry1Ac Bt rice Huahui No. 1 (GM crop) and its parental counterpart, Minghui63. We identified a total of 2579 quantifiable bacterial operational taxonomic units (OTUs). Many treatment-enriched microbial OTUs were identified, including 14 NonGM-enriched OTUs and 10 GM-enriched OTUs. OTUs belonging to the phyla Proteobacteria, Actinobacteria, Acidobacteria, Firmicutes, Nitrospirae, Chlorobi and GN04 were identified as statistically different in abundance between GM and the other two treatments. Compared with the different impacts of different rice varieties on microbiota, the impact of rice planting on microbiota is more obvious. Furthermore, Huahui No. 1 transgenic Bt rice had a greater impact on the rhizosphere bacterial communities than Minghui63. Early developmental stages of the transgenic Bt rice had a significant impact on many Bacillaceae communities. Soil chemical properties were not significantly altered by the presence of transgenic Bt rice. The peak concentration level of Bt protein products was detected during the seedling stage of transgenic Bt rice, which may be an intriguing factor for bacterial diversity variations. Based on these findings, we conclude that transgenic Bt rice has a significant impact on root-associated bacteria. This information may be leveraged in future environmental safety assessments of transgenic Bt rice varieties.

Effects of soil microbial ecology on ginsenoside accumulation in Panax ginseng across different cultivation years

The quality of ginseng is greatly influenced by ecological factors, especially soil factors. However, there are few reports on the association between the physicochemical properties of ginseng rhizosphere soil, enzyme activity, microbial bacterial community, and ginsenoside content accumulation in ginseng planted at different ages. In order to investigate the relationship between the soil microbial ecology of ginseng and the content of ginsenosides, this study compared the differences in the rhizosphere soil microecological environment and ginsenoside accumulation between ginseng planted for one, two, and three years, using soil without ginseng cultivation as a control. The results showed that monomer saponins Ro, Re, Rf, Rb1, and total ginsenoside content increased significantly (P < 0.05), while monomer saponins Rb2, Rd, Rh2, and Rg3 content decreased significantly in three-year-old ginseng, (P < 0.05). And soil pH, soil bulk weight and soil enzyme activity in ginseng rhizosphere soils altered the abundance and diversity of rhizosphere soil bacterial community. Redundancy analysis and Pearson correlation analysis revealed that soil pH, soil bulk density, soil urease, and protease were the primary factors influencing the bacterial community in the soil around roots of one, two, three years old ginseng. Co-occurrence network visualization analysis indicated that seven bacterial communities, Planctomycetota, Methylomirabilota, Verrucomicrobiota and Acidobacteriota at the phylum level, Vicinamibacterales_unclassified, Vicinamibacteraceae_unclassified and Candidatus_Udaeobacter at the genus level, had a significant effect on the synthesis of ginsenosides. In summary, soil urease, pH, and soil bulk density in agricultural soils can regulate soil physicochemical properties, soil enzymatic activities, and soil bacterial community abundance. These factors are key drivers of the ginseng soil microbial ecology in ginseng cultivation. These studies will provide a scientific basis for subsequent optimization of soil improvement techniques for cultivated ginseng in farmland by improving the soil microbial ecology.

Comparing the Potential of Silicon Nanoparticles and Conventional Silicon for Salinity Stress Alleviation in Soybean (Glycine max L.): Growth and Physiological Traits and Rhizosphere/Endophytic Bacterial Communities.

In this study, 20-day-old soybean plants were watered with 100 mL of 100 mM NaCl solution and sprayed with silica nanoparticles (SiO2 NPs) or potassium silicate every 3 days over 15 days, with a final dosage of 12 mg of SiO2 per plant. We assessed the alterations in the plant's growth and physiological traits, and the responses of bacterial microbiome within the leaf endosphere, rhizosphere, and root endosphere. The result showed that the type of silicon did not significantly impact most of the plant parameters. However, the bacterial communities within the leaf and root endospheres had a stronger response to SiO2 NPs treatment, showing enrichment of 24 and 13 microbial taxa, respectively, compared with the silicate treatment, which led to the enrichment of 9 and 8 taxonomic taxa, respectively. The rhizosphere bacterial communities were less sensitive to SiO2 NPs, enriching only 2 microbial clades, compared to the 8 clades enriched by silicate treatment. Furthermore, SiO2 NPs treatment enriched beneficial genera, such as Pseudomonas, Bacillus, and Variovorax in the leaf and root endosphere, likely enhancing plant growth and salinity stress resistance. These findings highlight the potential of SiO2 NPs for foliar application in sustainable farming by enhancing plant-microbe interactions to improve salinity tolerance.

Soil microorganisms play an important role in the detrimental impact of biodegradable microplastics on plants

Biodegradable plastics were developed to mitigate environmental pollution caused by conventional plastics. Research indicates that biodegradable microplastics still have effects on plants and microorganisms as their non-biodegradable counterparts, yet the effects on vegetable crops are not well-documented. Additionally, the function of soil microorganisms affected by biodegradable microplastics on the fate of microplastics remains unverified. In this study, Brassica chinensis was cultivated in soil previously incubated for one year with low-density polyethylene (LDPE-MPs) and poly (butylene adipate-co-terephthalate) microplastics (PBAT-MPs) at 0.05 % and 2 % concentrations. High concentrations of PBAT-MPs significantly reduced the biomass to 5.83 % of the control. The abundance of Methyloversatilis, IS-44, and UTCFX1 in the rhizosphere bacterial community increased significantly in the presence of PBAT-MPs. Moreover, these microplastics significantly enhanced soil enzyme activity. Incubation tests were performed with three PBAT plastic sheets to assess the function of the altered bacterial community in the soil of control (Control-soil) and soil treated with high concentrations of PBAT-MPs (PBAT-MPs-soil). Scanning Electron Microscopy and Atomic Transfer Microscopy (SEM/ATM) results confirmed enhanced PBAT degradation in the PBAT-MPs-soil. PICRUST2 analysis revealed that pathways related to substance degradation were upregulated in the PBAT-MPs-soil. Furthermore, a higher percentage of strains with PBAT-MPs-degrading ability was found in PBAT-MPs-soil. Our results confirm that PBAT-MPs significantly inhibit the growth of vegetable crops and that soil bacterial communities affected by PBAT-MPs are instrumental in degrading them.

Tomato domestication rather than subsequent breeding events reduces microbial associations related to phosphorus recovery

Legacy phosphorus (P) is a reservoir of sparingly available P, and its recovery could enhance sustainable use of nonrenewable mineral fertilizers. Domestication has affected P acquisition, but it is unknown if subsequent breeding efforts, like the Green Revolution (GR), had a similar effect. We examined how domestication and breeding events altered P acquisition by growing wild, traditional (pre-GR), and modern (post-GR) tomato in soil with legacy P but low bioavailable P. Wild tomatoes, particularly accession LA0716 (Solanum pennellii), heavily cultured rhizosphere P solubilizers, suggesting reliance on microbial associations to acquire P. Wild tomato also had a greater abundance of other putatively beneficial bacteria, including those that produce chelating agents and antibiotic compounds. Although wild tomatoes had a high abundance of these P solubilizers, they had lower relative biomass and greater P stress factor than traditional or modern tomato. Compared to wild tomato, domesticated tomato was more tolerant to P deficiency, and both cultivated groups had a similar rhizosphere bacterial community composition. Ultimately, this study suggests that while domestication changed tomato P recovery by reducing microbial associations, subsequent breeding processes have not further impacted microbial P acquisition mechanisms. Selecting microbial P-related traits that diminished with domestication may therefore increase legacy P solubilization.

Interactions between root and rhizosphere microbes mediate phosphorus acquisition in Pinus tabulaeformis

The Pinus tabulaeformis plantation is a major industrial forest in China. Phosphorus (P) deficiency seriously affected its growth and productivity. Root traits and rhizosphere microorganisms play essential roles in plant P acquisition. However, how root traits interact with microbial communities for P acquisition remains largely unclear. The responses of P. tabulaeformis root morphological characteristics, oxalic acid, soil P fractions and rhizosphere bacterial and fungal communities and potential functions to P addition were investigated using a pot experiment with three P levels (0, 20, and 80 mg P kg−1 soil). Additionally, we constructed a functional synthetic microbial community (SynCom) and verified their behaviors in promoting plant P acquisition and growth. P fertilization significantly increased the soil CaCl2-P, citrate-P, HCl-P and microbial biomass P (MBP) fractions and decreased oxalic acid, bacterial indicator genera in P deficiency and ectomycorrhizal fungi (ECM). Most of the bacterial indicator genera in P deficiency were negatively correlated with citrate-P and HCl-P but were positively correlated with alkaline phosphatase (ALP) and oxalic acid. According to structure equation models, soil total P could directly influence P acquisition efficiency and indirectly through its effects on ECM or by regulating oxalic acid, which regulated bacterial indicator genera in P deficiency and thus affected ALP production. Inoculation with SynCom significantly increased soil available P contents, ALP activities, root P concentrations and superoxide dismutase activities. Under low P conditions, P. tabulaeformis adopted a high carbon cost strategy for P acquisition, specifically moderating root secretion. Rhizosphere bacteria could accelerate soil P mobility and enhance the P absorption ability and systemic resistance of roots to alleviate P stress. This study provided the theoretical basis for the exploitation and utilization of microbial fertilizers, as well as for the cultivation and management of plantation.

Assisting nickel agromining using sustainable amendments

AbstractOne of the challenges of agromining is the adoption of more environmentally‐friendly solutions to improve plant biomass yields and Ni concentrations in plants. Here, we focused on four sustainable solutions for optimizing nickel phytoextraction by the hyperaccumulator Odontarrhena chalcidica: a biostimulant, another biostimulant/plant defense stimulator, artificial root exudates, and a biodegradable metal chelator. Their effects on the growth and physiology of O. chalcidica, on Ni phytoextraction capacity, on physicochemical soil characteristics, and on the diversity of rhizosphere and endophytic bacteria were compared to a conventional mineral fertilizer. A 5‐month pot experiment was carried out with O. chalcidica growing on an ultramafic soil. Element concentrations in both soil and plant were measured. Moreover, numerous compounds were analyzed (photosynthetic pigments, malondialdehyde, flavonoids, free amino acids, and starch). We also characterized rhizosphere and endophytic bacterial communities associated with this hyperaccumulator. Biostimulants appeared to be a promising way of improving Ni concentration in shoots and plant biomass production, and showed a positive effect on bacterial richness and diversity. In contrast, our experiments did not show that artificial exudates and mineral fertilizer had a positive effect on Ni phytoextraction. Finally, the biodegradable chelator had no significant effect. The use of sustainable amendments into a Ni agromining system improved both plant biomass and Ni yields, in comparison to mineral fertilization. Thus, improving agromining by replacing mineral fertilizers would be an eco‐efficient strategy.

The Contributions of Sub-Communities to the Assembly Process and Ecological Mechanisms of Bacterial Communities along the Cotton Soil–Root Continuum Niche Gradient

Soil microbes are crucial in shaping the root-associated microbial communities. In this study, we analyzed the effect of the soil–root niche gradient on the diversity, composition, and assembly of the bacterial community and co-occurrence network of two cotton varieties. The results revealed that the bacterial communities in cotton soil–root compartment niches exhibited a skewed species abundance distribution, dominated by abundant taxa showing a strong spatial specificity. The assembly processes of the rhizosphere bacterial communities were mainly driven by stochastic processes, dominated by the enrichment pattern and supplemented by the depletion pattern to recruit bacteria from the bulk soil, resulting in a more stable bacterial community. The assembly processes of the endosphere bacterial communities were determined by processes dominated by the depletion pattern and supplemented by the enrichment pattern to recruit species from the rhizosphere, resulting in a decrease in the stability and complexity of the community co-occurrence network. The compartment niche shaped the diversity of the bacterial communities, and the cotton variety genotype was an important source of diversity in bacterial communities within the compartment niche. We suggest that the moderate taxa contribute to significantly more changes in the diversity of the bacterial community than the rare and abundant taxa during the succession of bacterial communities in the cotton root–soil continuum.

Soil Bacterial Community of Medicinal Plant Rhizosphere in a Mediterranean System

Several attempts have been made to evaluate the abundance and distribution of the bacterial community in the rhizosphere of medicinal plants. Many describe information based on an estimation of the community structure and the effects of plant cover in determining microbial community composition. The ability of plants to specifically shape their microbial community in general and medicinal plants in particular is largely unknown. With the arrival of molecular biology, understanding the microbial community’s composition, diversity, and function became possible. We hypothesized that microbial communities associated with medicinal shrubs would differ from each other. To test this hypothesis, we characterized the soil microbial composition under each of five Mediterranean medicinal plants, differentiated by their medicinal use and ecophysiological adaptation, namely, Salvia fruticosa, Pistacia lentiscus, Myrtus communis, Origanum syriacum, and Teucrium capitatum, and an open-space bare soil between the plants, inhabiting natural ecosystems characterized by similar climatic conditions typical of a Mediterranean environment. The results demonstrated the importance of plant ecophysiological adaptations, which play an important role in determining microbial community composition and functional diversity. The intensity of a plant’s response to its surroundings can have either positive or negative effects that will determine the microbial community composition and interactions among the belowground parts. A total of 11 phyla, 21 orders, and 409 genera were found in the soil rhizosphere in the vicinity of the four plants and open space samples. The distinguishing attributes of each shrub trigger and stimulate the microbial community’s rhizosphere. This results in distinct patterns of bacterial diversity and functionality between the different shrubs and the control. The rhizosphere bacterial community composition differed between the plants in a PERMANOVA test, but there was little difference in terms of phyla and order relative abundances. This study shows how five medicinal plants, coexisting in a common habitat, impact the bacterial community. The noticeable shift in bacterial composition further supports our discovery that root exudates effectively govern the makeup of soil bacterial communities.

Diversity and growth-promoting characteristics of rhizosphere bacteria of three naturally growing plants at the sand iron ore restoration area in Qinghe County

It is a great challenge to restore northern mines after mining and achieve optimal results due to the extremely harsh environment and climate, as in Qinghe County of Xinjiang Province, China. Qinghe County has a climate of drought, cold, strong winds, and high altitude. After sand and iron mining, the soil in this area contains a large amount of sand and gravel with extremely low organic matter, nitrogen deficiency, and a high pH of 9.26. Our preliminary studies disclosed that only three plants, including Caligonum junceum, Atraphaxis virgata, and Melilotus albus Medic, can grow naturally in this environment without any artificial management. For effective ecology restoration, this study explored the mechanism of plant-microbial interaction and stress resistance in this environment. It was found that although the soil condition in the sand iron ore landfill area is extreme, the bacterial diversity remained high, with Shannon and Simpson indices reaching 9.135 and 0.994, respectively. The planting of three types of remediation plants did not significantly improve, or even decreased, the soil bacterial diversity index, but greatly changed the composition of dominant bacterial genera. Significant differences in the composition of rhizosphere soil bacterial communities among these three remediation plants were observed. Potential new bacterial species accounted for 9.8 %, and the proportion of unique genera reached 30 % or 50 %, respectively. Among all the isolated strains, 74 % had nitrogen fixation and other growth-promoting properties. In summary, the soil microbial community structure in this extreme environment is unique and diverse. The types of remediation plants play a major role in the composition of the rhizosphere bacterial community structure, and the recruited growth-promoting bacteria are diverse and functional. This study may offer valuable information for further studies in vegetation restoration and aid in ecology restoration, especially under extreme conditions.

Impacts of AlaAT3 transgenic poplar on rhizosphere soil chemical properties, enzyme activity, bacterial community, and metabolites under two nitrogen conditions

ABSTRACT Poplar stands as one of the primary afforestation trees globally. We successfully generated transgenic poplar trees characterized by enhanced biomass under identical nutrient conditions, through the overexpression of the pivotal nitrogen assimilation gene, pxAlaAT3. An environmental risk assessment was conducted for investigate the potential changes in rhizosphere soil associated with these overexpressing lines (OL). The results show that acid phosphatase activity was significantly altered under ammonium in OL compared to the wild-type control (WT), and a similar difference was observed for protease under nitrate. 16SrDNA sequencing indicated no significant divergence in rhizosphere soil microbial community diversity between WT and OL. Metabolomics analysis revealed that the OL caused minimal alterations in the metabolites of the rhizosphere soil, posing no potential harm to the environment. With these findings in mind, we anticipate that overexpressed plants will not adversely impact the surrounding soil environment.

Two microbes assisting Miscanthus floridulus in remediating multi-metal(loid)s-contaminated soil.

Miscanthus has good tolerance to multi-metal(loid)s and has received increasing attention in remediated studies of metal(loid)s-contaminated soil. In this study, we conducted phytoextraction techniques to investigate the synergic effects of remediation of multi-metal(loid)s-contaminated soil by Miscanthus floridulus (Lab.) and two plant growth-promoting bacteria (PGPB), TS8 and MR2, affiliated to Enterobacteriaceae. The results exhibited a decrease of arsenic (15.27-21.50%), cadmium (8.64-15.52%), plumbum (5.92-12.76%), and zinc (12.84-24.20%) except for copper contents in the soil in bacterial inoculation groups, indicating that MR2 and TS8 could enhance the remediation of metal(loid)s. Moreover, increased fresh/dry weight and height indicated that inoculated bacteria could promote Miscanthus growth. Although the activities of antioxidant enzymes and the content of chlorophyll in the overground tissues showed no significant increase or even decrease, the activities of antioxidant enzymes in the underground tissues and soil were elevated by 48.95-354.17%, available P by 19.07-23.02%, and available K by 15.34-17.79% (p < 0.05). Bacterial inoculants could also decrease the soil pH. High-throughput sequencing analysis showed that the bacterial inoculant affected the rhizosphere bacterial community and reduced community diversity, but the relative abundance of some PGPB was found to increase. Phylogenetic molecular ecological networks indicated that bacterial inoculants reduced interactions between rhizosphere bacteria and thereby led to a simpler network structure but increased the proportion of positive-correlation links and enhanced the metabiosis and symbiosis of those bacteria. Spearman's test showed that OTUs affiliated with Enterobacteriaceae and soil nutrients were critical for metal(loid) remediation and Miscanthus growth. The results of this study provide a basis for the synergic remediation of multi-metal(loid)s-contaminated soils by Miscanthus and PGPB and provide a reference for the subsequent regulation of Miscanthus remediation efficiency by the other PGPB or critical bacteria.

Phthalate ester (PAEs) accumulation in wheat tissues and dynamic changes of rhizosphere microorganisms in the field with plastic-film residue

Phthalates acid esters (PAEs) have accumulated in soil and crops like wheat as a result of the widespread usage of plastic films. It is yet unclear, nevertheless, how these dynamic variations in PAE accumulation in wheat tissues relate to rhizosphere bacteria in the field. In this work, a field root-bag experiment was conducted to examine the changes of PAEs accumulation in the rhizosphere soil and wheat tissues under film residue conditions at four different growth stages of wheat, and to clarify the roles played by the microbial community in the alterations. Results showed that the plastic film residues significantly increased the concentrations of PAEs in soils, wheat roots, straw and grains. The maximum ΣPAEs concentration in soils and different wheat tissues appeared at the maturity, with the ΣPAEs concentration of 1.57 mg kg−1, 4.77 mg kg−1, 5.21 mg kg−1, 1.81 mg kg−1 for rhizosphere soils, wheat roots, straw and grains, respectively. The plastic film residues significantly changed the functions and components of the bacterial community, increased the stochastic processes of the bacterial community assembly, and reduced the complexity and stability of the bacterial network. In addition, the present study identified some bacteria associated with plastic film residues and PAEs degradation in key-stone taxa, and their relative abundances were positive related to the ΣPAEs concentration in soils. The PAEs content and key-stone taxa in rhizosphere soil play a crucial role in the formation of rhizosphere soil bacterial communities. This field study provides valuable information for better understanding the role of microorganisms in the complex system consisting of film residue, soil and crops.

Improved bacterial composition and co-occurrence patterns of rhizosphere increased nutrient uptake and grain yield through cultivars mixtures in maize

Crop diversification contributes to agricultural productivity and resources efficient utilization. However, whether cultivar mixtures in maize affects soil bacterial community, nutrient uptake, plant growth and yield remains unknown. A two-year lysimetric experiment was conducted using two maize cultivars (LY16 and JS501) with different root system architectures planted in monoculture or in mixture under normal fertilization (NF), reduced fertilization (RF) or no addition of fertilizer (CK) and was assessed at the silking stages. Cultivar mixtures and monoculture of LY16 had higher shoot biomass, nutrient uptake and total root length at silking stage, and grain yield than monoculture of JS501 under NF and RF conditions. Under NF and RF conditions, cultivar mixtures and monoculture of LY16 led to an increase in bacterial diversity, significant changes in community structure, and a high abundance of Bacteroidia and biomarkers of Chitinophagaceae and Saprospiraceae (Bacteroidia). Cultivar mixtures showed specific responses from modules of the rhizosphere bacterial community co-occurrence network, and the relative abundance of keystone taxa of cultivar mixtures was higher than that of monoculture of JS501. The keystone taxa had a broad and significant positive correlation with plant nutrient accumulation and grain yield. Cultivar mixtures showed similar assembly processes of Bacteroidia with monoculture of LY16, and the increased abundance of Chitinophagaceae may lead to a healthy rhizosphere bacterial community. Overall, our findings indicate that cultivar mixtures significantly affects the assembly and composition of the rhizosphere bacterial community, and thus benefits plant nutrient acquisition and plant growth. These findings could deepen our understanding of the facilitating effect of rhizosphere functional microbial community (e.g. plant nutrition uptake or immunity)of cultivar mixtures.

Impacts of nano-acetamiprid pesticide on faba bean root metabolic response and soil health

The associated benefits and potential environmental risks of nanopesticides on plant and soil health, particularly in comparison with traditional pesticides, have not been systematically elucidated. Herein, we investigated the impacts of the as-synthesized nano-acetamiprid (Nano-Ace, 20 nm) at low (10 mg/L), medium (50 mg/L), high (100 mg/L) doses and the corresponding high commercial acetamiprid (Ace, 100 mg/L) on the physiological and metabolic response of faba bean (Vicia faba L.) plants, as well as on rhizosphere bacterial communities and functions over short-, medium- and long-term exposures. Overall, Nano-Ace exposure contributed to basic metabolic pathways (e.g., flavonoids, amino acids, TCA cycle intermediate, etc.) in faba bean roots across the whole exposure period. Moreover, Nano-Ace exposure enriched rhizosphere beneficial bacteria (e.g., Streptomyces (420.7%), Pseudomonas (33.8%), Flavobacterium (23.3%)) and suppressed pathogenic bacteria (e.g., Acidovorax (44.5%)). Additionally, Nano-Ace exposure showed a trend of low promotion and high inhibition of soil enzyme activities (e.g., invertase, urease, arylsulfatase, alkaline phosphatase) involved in soil C, N, S, and P cycling, while the inhibition was generally weaker than that of conventional Ace. Altogether, this study indicated that the redox-responsive nano-acetamiprid pesticide possessed high safety for host plants and soil health.

Relationships between rhizosphere microbial communities, soil abiotic properties and root trait variation within a pine species

Abstract Rhizosphere microbes play important roles in plant performance and ecosystem functioning. It is becoming increasingly clear that rhizosphere communities vary with soil properties and variation in root traits among plant species. However, less is known about whether and how variation in root traits within plant species influences the rhizosphere microbial communities. We evaluated the intraspecific root traits variation and explored their associations with bacterial and fungal communities in rhizosphere by focusing on an ectomycorrhizal tree species, that is Pinus massoniana, in 22 sites in subtropical China. The first dimension of the principal component analysis on root traits revealed evidence for the ‘conservation’ gradient of the root economics space. Overall, root traits explained more variation in fungal communities than in bacterial communities in the rhizosphere. Functional composition of rhizosphere microbial communities changed significantly along the ‘conservation’ gradient, with fast‐growing copiotrophic bacteria and symbiotic ectomycorrhizal fungi were significantly enriched on the ‘acquisition’ side, while slow‐growing oligotrophic bacteria were significantly enriched on the ‘conservation’ side of the gradient. Synthesis: Our study demonstrates that intraspecific variation in plant roots significantly influence rhizosphere microbial communities, which in turn can influence plant nutrition and therefore plant performance within the community.

Temporal dynamics of rhizosphere bacterial community in the Robinia pseudoacacia–Mesorhizobium loti symbiotic system for remediation of cadmium-contaminated soils

Rhizobia can be used to enhance the phytoremediation of cadmium-contaminated soils by legumes, and a symbiotic nitrogen-fixing system has been shown to reshape the structure of rhizosphere microbiota in a mining area. However, little is known about the temporal dynamics and possible roles of rhizosphere microbiota in rhizobia-enhanced phytoremediation. A rhizobox experiment was conducted using Robinia pseudoacacia alone or inoculated with Mesorhizobium loti HZ76 under greenhouse conditions. Rhizosphere bacterial communities were characterized over a 90-day period at three levels of cadmium (determined by the distance from the mining area; uncontaminated: 0.08 mg kg−1, contaminated: 0.39 and 1.72 mg kg−1). Rhizobial inoculation considerably improved the phytoremediation efficiency, such as increasing plant biomass and cadmium accumulation by approximately two times at 90 days after planting. The bacterial community structure shifted remarkably with increasing duration of remediation. Bacterial diversity exhibited a decreasing trend in the first 60 days and then recovered at 90 days. Despite having no discernible impact on the bacterial community structure in the rhizosphere, rhizobial inoculation enabled the enrichment of potentially beneficial taxa (e.g., Microvirga, Mesorhizobium, Diaphorobacter, Steroidobacter, Brevundimonas) with resistance to heavy metals and involvement in nutrient cycling. The bacterial co-occurrence networks of low and high cadmium-contaminated soils showed the highest average degree and clustering coefficient at 60 and 30 days, respectively. Some bacterial taxa involved in nitrogen fixation (e.g., Rhizobacter, Mesorhizobium), denitrification (e.g., Niastella, Nitrospira), and carbon turnover (e.g., Solirubrobacter, Brevundimonas) were found to be potential keystone taxa. The collective findings clarify possible mechanisms by which rhizosphere bacteria participate in phytoremediation of cadmium-contaminated mining soils using the R. pseudoacacia–M. loti symbiotic system.

Comparison of Rhizosphere Bacterial Communities of Pinus squamata, a Plant Species with Extremely Small Populations (PSESP) in Different Conservation Sites.

Pinus squamata is one of the most threatened conifer species in the world. It is endemic to northeastern Yunnan Province, China, and has been prioritized as a Plant Species with Extremely Small Populations (PSESP). The integrated study of soil properties and rhizosphere bacteria can assist conservation to understand the required conditions for the protection and survival of rare and endangered species. However, differences between the rhizospheric bacterial communities found in the soil surrounding P. squamata at different conservation sites remain unclear. In this study, Samples were collected from wild, ex situ, and reintroduced sites. Bacterial communities in different conservation sites of P. squamata rhizosphere soils were compared using Illumina sequencing. The soil physicochemical properties were determined, the relationships between the bacterial communities and soil physicochemical factors were analyzed, and the potential bacterial ecological functions were predicted. The reintroduced site Qiaojia (RQ) had the highest richness and diversity of bacterial community. Actinobacteria, Proteobacteria, and Acidobacteriota were the dominant phyla, and Bradyrhizobium, Mycobacterium, Acidothermus were the most abundant genera. Samples were scattered (R = 0.93, p = 0.001), indicating significant difference between the different conservation sites. The abundance of Mycobacterium differed between sites (0.01 < p ≤ 0.05), and the relative abundances of Bradyrhizobium and Acidothermus differed significantly among different sites (0.001 < p ≤ 0.01). Soil total potassium (TK) and available nitrogen (AN) were the main factors driving bacterial community at the phylum level (0.01 < p ≤ 0.05). This study generated the first insights into the diversity, compositions, and potential functions of bacterial communities associated with the rhizosphere soils of P. squamata in different conservation sites and provides a foundation to assess the effect of conservation based on bacterial diversity and plant growth-promoting rhizobacteria (PGPR) to guide future research into the conservation of P. squamata.
